Randy Wayne est né et a grandi à Moore, Oklahoma.
Il est surtout célèbre pour son rôle de Luke Duke dans le film The Dukes of Hazzard: The Beginning. Depuis, il a joué dans de nombreux films indépendants, y compris Dream Boy, Grizzly Park. Il a également joué Michael dans The Haunting of Molly Hartley, The Last Hurrah, et Ghost Town (un film de 2009 Syfy horreur anglo-normandes, à ne pas confondre avec le film de 2008 Ricky Gervais du même nom). Il a joué dans le rôle principal de Jake Taylor To Save a Life, qui est sorti en salles le 22 Janvier 2010.
